Breeding of Teicoplanin High-yield Strain from Actinomycetes and Optimization of Fermentation Conditions

In this issue, we recommend an article published by the research team of Ma Jie, a teacher of Hebei Industrial microbial metabolic engineering research center, in the well-known journal China Journal of antibiotics:Breeding of Teicoplanin High-yield Strain from Actinomycetes and  Optimization of Fermentation Conditions.


Abstract

To obtain a new strain with genetic stability and higher yield of teicoplain. Methods A new strain with stable heredity and high yield was obtained by the atmospheric pressure room temperature plasma (ARTP) mutagenesis treatment using a teicoplanin-producing bacteria Antinoplanes teichomycetics T19 as the starting strain. Fermentation conditions such as rotation speed, inoculation amount, temperature, and pH were optimized to improve the yield of teicoplanin. Results The fermentation titer of the mutagenized strain was 42% higher than that of the original strain. When rotation speed was 220 r/min, inoculation amount was 2%, temperature was 28℃, and pH was 6.5, the fermentation titer of this strain was 80% higher than that of the original strain. Conclusion The ARTP mutagenesis technology could be effectively used for the mutagenesis and breeding of the Actinoplanes to increase the yield of teicoplanin, which laid a foundation for its industrial production.
